Guitar mini game?

For A Game i am making i'm trying to add a mini game like guitar hero or something else where you have to hit something and can hit the wrong one or do it at the wrong time. i just don't know how I Can Make it where if an object reaches a certian point it get's to a game over or something like that.....

Any Help Would be nice.
