When I first saw RMVX I was slightly disappointed with the tilesets (the way they looked) but over time I have started to like them. The simple to use "Quick Event Creation Tool" (for things like Inns, which could take awhile before) will speed up the game making process, but not take the fun out of it. All the menus and options look, simply, amazing! I find the in-game layout to be simply better, and the database also looks less complicated with more options.
According to the features page of the Enterbrains site, there are in fact no more layers in the sence me and you know them. However, you can simply "pile" tiles ontop of others so you can use as many layers as needed to get the job done. I'm not overly sure if this is a step forward or backward though. Perhaps one of the most disappointing things about it all is that they made it too basic? If it is at all possible, of course. But I hope it still is as fun as ever to create the games rather then just play them. And speaking of playing them, more games should be finished now thanks to the quicker game making process.
RGSS2 also could provide even more possiblities! I'm not sure what the exact changes are from RGSS are but if anything, it will probably be able to handle more complex scripts to allow for many new features for everyones games.
Built-in transport & Dash scripts are sure to go down a treat aswell!
Deeply looking forward to the RMVX release as he upgraded to Vista and can't get RMXP to work,

PS. I didn't oversell it, did I?
