Title: How to make things done in paint transparent [ I need help!]

The title says it all; how do I make the backgrounds of pictures edited in Paint transparent so I can use them in programs like rmvx and rmxp? I was trying to change the title used for Moghunter's animated menu script and then I came across this problem. Will I need Photoshop?

What you do is you make the background all one color and save it as a .png file. You then open Resource Manager of the RPG maker and import the picture you want. At that point it will ask you what color you want as transparent and what color you want as semi-transparent.
